Disease Information: Mycobacterium Leprae is a slow-growing bacteria that causes Leprosy, which is also known as Hansen's Disease. Leprosy is a human disease that affects the skin, peripheral nerves, eyes, and mucosa of the upper respiratory tract. The bacteria causes numbness, nerve damage, and gradual tissue decay and disfigurement of the limbs. The nerve damage from the disease can lead paralysis and crippled hands and feet. The signs and symptoms of leprosy mainly affect the skin, nerves, and mucous membranes. Some signs are discolored skin patches, loss of eyebrow hair or eyelashes, muscle weakness or paralysis, enlarged nerves, etc.

Essentiality: Malate synthase catalyzes the irreversible condensation of acetyl-CoA with glyoxylate which forms (S)-malate. Also, the enzyme plays a role in the glyoxylate cycle, which leads to the loss of CO(2).

Assayed by adding cell extracts to H2O2 in K2HPO4. O-dianisidine was used for peroxidase and was added to a phosphate buffer and H2O2 in deionized water.

Materials and Reagents:


The enzyme catalyses the irreversible condensation of acetyl-CoA with glyoxylate to form (S)-malate. Among other functions, the enzyme participates in the glyoxylate cycle, a modified version of the TCA cycle that bypasses steps that lead to a loss of CO2.
